They will absolutely keep the same sound signature…though you’ll need to make sure to save the extra foam layer that’s underneath the stock M40X pads and put them underneath these as well, as that foam piece is part of the sound tuning on the 40's.

Comfort improvements aren’t dramatic, but yes, the M50 pads are a bit softer and more conforming to the head than the stock M40X pads, so you should notice an improvement. The M40’s also benefit from stretching/breaking in, as I’ve found they have the highest clamping force of the M-series lineup.
