I test every headset and headphone with my glasses on, only removing them for a moment to see if they have any significant impact on the seal and sound quality. Any comfort claims I make refer to listening with my glasses on, unless I specifically state otherwise.

The Opus is a very comfy headphone on my head whether I'm wearing glasses or not. It clamps a little bit stronger than the average headphone, so that might bother you especially for the first day or so, but the pads are very soft and the comfort design is generally excellent.
